2. Abstract

RNA interference (RNAi)는 double stranded RNA (dsRNA).로 생기는 gene silencing process 이다. double-stranded RNA (dsRNA) 을 cell 내로 주입할 때 RNA-based gene silencing (RBGS)가 일어나는데 dsRNA-specific endonuclease, Dicer로 인하여 double stranded short interfering RNAs (siRNAs)가 만들어진다. siRNA 와 효소들의 복합체인 RNA-induced silencing complex (RISC) 는 mRNA 의 특정 장소로 이동하여 endonuclease and exonuclease activities를 가지고 target mRNA를 degrade하거나 translational suppresion을 일으킨다. double stranded RNA 는 single strand RNA보다 강력하고 specific하게 interference를 일으키며 interference의 영향은 자손들에게 까지도 전해지고 아주 적은 양으로도 interference process에 catalytic amplification 이 될 수 있다. 많은 연구자들은 siRNAs 가 endogenous and exogenous transposons and retroelements에 대하여 defense 한다고 보고 있으며 siRNA가 immune system에 강력한 therapy application이자 experimental tool이 될 것이라 생각한다.
