The objective of this study was to investigate the anti-obesity activity of the ethanol extract of whole barley (WBE) roasted at 150, 190, and 230oC on 3T3-L1 adipocytes and oleic acid (OA)-induced HepG2 hepatocytes. The WBE inhibited fat accumulation in 3T3-L1 and OA-induced HepG2 as its roasting temperature increased. At 100 μg/mL of 190oC WBE, the expression of genes related to adipogenic differentiation in 3T3-L1, such as pparγ, ap2, scd1, and acc1, was reduced by 21, 68, 39, and 63%, respectively, compared to that in adipocytes. Likewise, 75 μg/mL of 190oC WBE significantly inhibited the expression of transcription factors including pparγ, c/ebp-α, and srebp-1c in OA-induced HepG2 (p<0.05). In 3T3-L1, at 75 μg/mL of 190oC WBE, the protein levels of C/EBP-α, SREBP-1C, AP2, and SCD1, decreased by 31, 24, 45, and 52%, respectively. Overall, 190oC WBE inhibited fat accumulation in 3T3-L1 and HepG2, showing its potential use as a natural anti-obesity functional material.
