ε-Caprolactam 이용성(利用性) 세균(細菌)의 분리(分離) 및 그 성질(性質)

초록

대구(大邱) 신천(新川) 하상(河床) 오니(汚泥)로부터 -caprolactam이용성(利用性)이 우수한 균(菌)을 분리(分離)하여 Arthrobacter globiformis N-2-1로 동정(同定)했다. Arthrobacter globiformis N-2-1의 배양(培養)을 위한 최적배지(最適培地)의 조성(組成)은 -caprolactam 0.4%, 0.2%, 0.05%, 0.02%, 0.01%, yeast extract 0.05%이었다. 이 균(菌)의 생육(生育)을 위한 최적(最適) pH는 7.0, 온도(溫度)는 부근(附近)이었다. 본(本) 균(菌)은 무기염 배지(培地)에서 yeast extract를 가(加)하지 않고서도 -caprolactam을 잘 분해(分解)하여 증식(增殖)할 수 있었으나, yeast extract를 가(加)하면 생육(生育)이 더욱 촉진되었다. 또한 본(本) 균(菌)은 대부분의 당(糖)을 잘 이용(利用)할 수 있었고 유기산(有機酸)으로는 -ketoglutarate, adipate, p-hydroxybenzoate는 잘 이용(利用)하였으나 gluconate는 잘 이용(利用)할 수 없었다. 그리고 amino acid는 대부분 잘 이용(利用)하는 것으로 나타났다. 0.4%의 -caprolactam을 함유한 배지(培地)에서 60시간(時間) 배양(培養)으로 -caprolactam을 완전히 분해(分解)할 수 있었다. 분리균(分離菌) N-2-1에 의한 -caprolactam의 분해(分解) 생성물(生成物)은 -aminocaproic acid로 확인되었다.

영어초록

A bacterium which utilizes -caprolactam as a sole source of carbon and nitrogen was isolated from sludge of Shinchun river in Taegu and identified as Arthrobacter globiformis N-2-l. The growth medium for the optimum culture condition was composed of 0.4% -caprolactam, 0.02% , 0.05% , 0.02% , 0.01% and 0.05% yeast extract. The optimum pH and temperature for growth were 7.0 and respectively. The bacterial growth on the -caprolactam medium did not require any other organic nitrogen source such as yeast extract, although it was remarkably stimulated by the yeast extract. The bacteria utilized wide range of sugars and organic acids such as -ketoglutarate, adipate and P-hydroxybenzoate. The bacteria could use all kind of amino acids, -Caprolactam in the medium was consumed completely in the timecourse culture at for 60 hr on the shaker by the bacteria. Decomposition product of -caprolactam by Arthrobacter globiformis N-2-1 was -aminocaproic acid.
